Le laboratoire Immunologie-Immunopathologie-Immunotherapie (i3), créé en 2009, est consacré à l’immunologie des systèmes translationnelle. Il a comme objectif heuristique de contribuer à l’avancement des connaissances fondamentales en immunologie et comme objectif appliqué de développer des immunothérapies innovantes contre les maladies auto-immunes & inflammatoires et l’allergie. Ainsi, i3 met en œuvre des approches de biologie des systèmes pour analyser la complexité des interactions cellulaires et des régulations au cours des réponses immunitaires et en thérapeutique. i3 maîtrise les analyses de données complexes en cytomique, génétique, metagénomique microbienne, transcriptomique et protéomique, et a une expertise particulière pour l’analyse du répertoire TCR des lymphocytes T. i3 réunit des immunologistes, des cliniciens, des bioinformaticiens, des mathématiciens, des informaticiens et des statisticiens qui apportent les compétences multidisciplinaires nécessaires à son ambition.

Clinical immunology & Biotherapy (CiB)
The CiB theme aims to (i) explore the immune system in health and diseases and (ii) to develop and validate new immunotherapies in humans. This theme associates a clinical immunopathology component and a more fundamental component on Treg biology. CiB has a recognized expertise in translational immunology; it assembles researchers with scientific expertise in cellular immunology, and more specifically in Treg, and practicing medical doctors with hospital activities in the field of immunological diseases. Importantly, CiB’s work is part of large and well-funded structures and projects, including the i2B department, the Transimmunom LabEx, the RHU iMAP and the ERC TRiPoD.

The EiB themes are to (i) explore the immune system in physiological and pathological contexts and (ii) develop and validate new immunotherapies in mice. EiB has a recognized expertise in Treg biology and vaccination. Exploiting our experience in vaccines against infectious diseases23,24, we developed innovative immunotherapies with the specific aim of inducing immune tolerance and applying the strategies to treat AIDs and allergy. Two different approaches were developed: (i) one based on Treg-mediated therapies, (ii) the other based on antigen-specific tolerogenic vaccination, for which proof of concept was recently achieved (cf patents). Our research was mainly based on pre-clinical studies focusing on models of immune-mediated disorders, notably allergies and AIDs, and supports the “pre-clinical development” team of the Clinical Investigation Center in Biotherapy and Immunology (CIC-BTi) located at Pitié-Salpêtrière Hospital.

Immunologie intégrative
i2 expertise is in developing systems immunology approaches to study T cell differentiation and dynamics in physiological and pathological situations relying on statistical and dynamic computer modelling. Integration of multi-scale and multi-level data (flow cytometry, transcriptome, proteome, microbiome & repertoire) allows modelling the dynamics and repertoire diversity of lymphocyte populations as well as analyzing their gene expression.
